Posh took an early lead before going down 3-2 at the Madejski Stadium on Tuesday night.
Tommy Rowe, after missing an earlier chance put the visitors ahead on 16 minutes, before Simon Church equalised inside half an hour.
1-1 at the break, Adam Le Fondre (77) and Hal Robson-Kanu (80) pushed Reading into a comfortable lead.
Posh flirted with another comeback in the final minute of normal time as Emile Sinclair hit Posh's second, but there was no more in the tank on this occasion.
Chairman Darragh MacAnthony commented on Twitter: "Thought we played fantastic for 65 mins and then ran out of steam big time.
